Genes affected
DNAH11 (HGNC:2942): (dynein axonemal heavy chain 11) This gene encodes a ciliary outer dynein arm protein and is a member of the dynein heavy chain family. It is a microtubule-dependent motor ATPase and has been reported to be involved in the movement of respiratory cilia. Mutations in this gene have been implicated in causing Kartagener Syndrome (a combination of situs inversus totalis and Primary Ciliary Dyskinesia (PCD), also called Immotile Cilia Syndrome 1 (ICS1)) and male sterility. The p.E2883K variant (also known as c.8647G>A), located in coding exon 52 of the DNAH11 gene, results from a G to A substitution at nucleotide position 8647. The glutamic acid at codon 2883 is replaced by lysine, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear. -

p.Glu2883Lys in exon 52 of DNAH11: This variant is not expected to have clinical significance due to a lack of conservation across species, including mammals. O f note, >20 species, including the opposum, tasmanian devil, wallaby and platypu s have a lysine (Lys) at this position. In addition, computational prediction to ols do not suggest a high likelihood of impact to the protein. It has been ident ified in 1/66670 European chromosomes by the Exome Aggregation Consortium (ExAC, http://exac.broadinstitute.org; dbSNP rs752605468). -
