This variant is reported in 0.023% of alleles in individuals of South Asian descent in gnomAD, which is more common than expected for an undocumented cause of disease. In ClinVar, this variant has conflicting interpretations including likely benign and uncertain significance (https://www.ncbi.nlm.nih.gov/clinvar/variation/477102/). The aspartic acid at codon 797 is replaced by glycine, an amino acid with similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -
