This variant is only observed on two alleles in the Genome Aggregation Database (v2.1.1), indicating it is not a common polymorphism. However, an alternative change at this amino acid location, (c.982C>T, p.Arg328Trp), has been reported in multiple individuals from a single family with malignant hyperthermia (Loke 2003). Four of five in-silico tools predict a damaging effect of the variant on protein function. The variant allele was found at a frequency of 8e-06 in 251392 control chromosomes. The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. To our knowledge, no occurrence of c.983G>A in individuals affected with Congenital Multicore Myopathy With External Ophthalmoplegia and no experimental evidence demonstrating its impact on protein function have been reported. ClinVar contains an entry for this variant (Variation ID: 559646). Based on the evidence outlined above, the variant was classified as uncertain significance. -
