Variant summary
Our verdict is Uncertain significance. The variant received 1 ACMG points: 1P and 0B. PP2
The NM_000157.4(GBA1):c.104C>T(p.Ser35Leu) variant causes a missense change involving the alteration of a non-conserved nucleotide. The variant allele was found at a frequency of 0.0000155 in 1,613,252 control chromosomes in the GnomAD database, with no homozygous occurrence. In-silico tool predicts a benign outcome for this variant. Variant has been reported in ClinVar as Uncertain significance (★★). Another variant affecting the same amino acid position, but resulting in a different missense (i.e. S35P) has been classified as Uncertain significance.

not specified Uncertain:1
The c.104C>T (p.S35L) alteration is located in exon 3 (coding exon 2) of the GBA gene. This alteration results from a C to T substitution at nucleotide position 104, causing the serine (S) at amino acid position 35 to be replaced by a leucine (L). Based on insufficient or conflicting evidence, the clinical significance of this alteration remains unclear. -

Gaucher disease Uncertain:1
The p.Ser35Leu variant in GBA has not been previously reported in individuals with Gaucher disease but has been identified in 0.004% (4/113450) of European (non-Finnish) chromosomes and 0.003% (1/30616) of South Asian chromosomes by the Genome Aggregation Database (gnomAD, http://gnomad.broadinstitute.org; dbSNP rs757041827). Although this variant has been seen in the general population, its frequency is low enough to be consistent with a recessive carrier frequency. This variant has also been reported in ClinVar (VariationID: 522661) as likely pathogenic by the Shahid Beheshti University of Medical Sciences. Computational prediction tools and conservation analyses do not provide strong support for or against an impact to the protein. In summary, the clinical significance of the p.Ser35Leu variant is uncertain. ACMG/AMP Criteria applied: PM2 (Richards 2015). -
