Intraflagellar transport is involved in the genesis, resorption and signaling of primary cilia. The primary cilium is a microtubule-based sensory organelle at the surface of most quiescent mammalian cells, that receives signals from its environment, such as the flow of fluid, light or odors, and transduces those signals to the nucleus. The c.4318C>T (p.R1440C) alteration is located in exon 31 (coding exon 29) of the IFT140 gene. This alteration results from a C to T substitution at nucleotide position 4318, causing the arginine (R) at amino acid position 1440 to be replaced by a cysteine (C). Based on insufficient or conflicting evidence, the clinical significance of this alteration remains unclear. -

This variant is present in population databases (rs758535401, gnomAD 0.02%). This variant has not been reported in the literature in individuals affected with IFT140-related conditions. ClinVar contains an entry for this variant (Variation ID: 465320).
