Genes affected
CRB1 (HGNC:2343): (crumbs cell polarity complex component 1) This gene encodes a protein which is similar to the Drosophila crumbs protein and localizes to the inner segment of mammalian photoreceptors. In Drosophila crumbs localizes to the stalk of the fly photoreceptor and may be a component of the molecular scaffold that controls proper development of polarity in the eye. Mutations in this gene are associated with a severe form of retinitis pigmentosa, RP12, and with Leber congenital amaurosis. Alternate splicing results in multiple transcript variants, some protein coding and some non-protein coding.[provided by RefSeq, Apr 2012]

Likely pathogenic, criteria provided, single submitterclinical testingGeneDxApr 03, 2018The C195F variant in the CRB1 gene has been reported previously in association with autosomal recessive CRB1-related disorder, in affected individuals who were compound heterozygous for the C195F variant and another pathogenic variant (den Hollander et al., 2004; Morarji et al., 2016). The C195F variant is not observed at a significant frequency in large population cohorts (Lek et al., 2016). The C195F variant is a non-conservative amino acid substitution, which is likely to impact secondary protein structure as these residues differ in polarity, charge, size and/or other properties. In-silico analyses, including protein predictors and evolutionary conservation, support a deleterious effect. We interpret C195F as a likely pathogenic variant. -

Pathogenic, criteria provided, single submitterclinical testingInvitaeDec 19, 2023This sequence change replaces cysteine, which is neutral and slightly polar, with phenylalanine, which is neutral and non-polar, at codon 195 of the CRB1 protein (p.Cys195Phe). This variant is present in population databases (rs764256655, gnomAD 0.004%). This missense change has been observed in individual(s) with CRB1-related disease (PMID: 15459956, 26914788, 27096895, 29391521). In at least one individual the data is consistent with being in trans (on the opposite chromosome) from a pathogenic variant. ClinVar contains an entry for this variant (Variation ID: 265083). Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is expected to disrupt CRB1 protein function with a positive predictive value of 95%. For these reasons, this variant has been classified as Pathogenic. -
