The FBN2 c.6598A>G variant is predicted to result in the amino acid substitution p.Met2200Val. To our knowledge, this variant has not been reported in the literature. This variant is reported in 0.0056% of alleles in individuals of Latino descent in gnomAD (http://gnomad.broadinstitute.org/variant/5-127624858-T-C). At this time, the clinical significance of this variant is uncertain due to the absence of conclusive functional and genetic evidence.

The p.M2200V variant (also known as c.6598A>G), located in coding exon 52 of the FBN2 gene, results from an A to G substitution at nucleotide position 6598. The methionine at codon 2200 is replaced by valine, an amino acid with highly similar properties. This amino acid position is well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.

Significance:Uncertain significance

Collection Method:clinical testing

The M2200V variant has not been published as a mutation, nor has it been reported as a benign polymorphism to our knowledge. The M2200V variant was not observed in approximately 6,500 individuals of European and African American ancestry in the NHLBI Exome Sequencing Project, indicating it is not a common benign variant in these populations. This substitution occurs at a position that is conserved across species. However, in silico analysis is inconsistent in its predictions as to whether or not the variant is damaging to the protein structure/function. The M2200V variant is a conservative amino acid substitution, which is not likely to impact secondary protein structure as these residues share similar properties. Additionally, no missense mutations in nearby residues have been reported in the Human Gene Mutation Database in association with Marfan syndrome. Therefore, based on the currently available information, it is unclear whether this variant is a pathogenic mutation or a rare benign variant.
