Serine palmitoyltransferase, which consists of two different subunits, is the key enzyme in sphingolipid biosynthesis. It catalyzes the pyridoxal-5-prime-phosphate-dependent condensation of L-serine and palmitoyl-CoA to 3-oxosphinganine. The alanine at codon 332 is replaced by serine, an amino acid with similar properties. This variant was detected in an individual with adult-onset spasticity; however, clinical details were limited (Ngo KJ et al. Hum Mutat, 2020 02;41:487-501). This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. - |
