The p.R187C variant (also known as c.559C>T), located in coding exon 4 of the PRX gene, results from a C to T substitution at nucleotide position 559. The arginine at codon 187 is replaced by cysteine, an amino acid with highly dissimilar properties. This variant was detected in a 42 year old female patient with CMT1C who was also found to have a heterozygous pathogenic variant in the LITAF gene (Khosa S et al. Cureus, 2020 Jun;12:e8517). This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -
Charcot-Marie-Tooth disease type 4 Uncertain:1
This sequence change replaces arginine, which is basic and polar, with cysteine, which is neutral and slightly polar, at codon 187 of the PRX protein (p.Arg187Cys). The frequency data for this variant in the population databases is considered unreliable, as metrics indicate poor data quality at this position in the gnomAD database. This variant has not been reported in the literature in individuals affected with PRX-related conditions. ClinVar contains an entry for this variant (Variation ID: 572807).
