The c.501-3C>A intronic variant results from a C to A substitution 3 nucleotides upstream from coding exon 5 in the RB1 gene. This variant was reported in individual(s) with retinoblastoma (Ambry internal data). This nucleotide position is not well conserved in available vertebrate species. In silico splice site analysis predicts that this alteration may weaken the native splice acceptor site; however, direct evidence is insufficient at this time (Ambry internal data). Based on the available evidence, the clinical significance of this variant remains unclear. -
