TGFBR2 (HGNC:11773): (transforming growth factor beta receptor 2) The protein encoded by this gene is a transmembrane protein that has a protein kinase domain, forms a heterodimeric complex with TGF-beta receptor type-1, and binds TGF-beta. This receptor/ligand complex phosphorylates proteins, which then enter the nucleus and regulate the transcription of genes related to cell proliferation, cell cycle arrest, wound healing, immunosuppression, and tumorigenesis. Mutations in this gene have been associated with Marfan Syndrome, Loeys-Deitz Aortic Aneurysm Syndrome, and the development of various types of tumors. Algorithms developed to predict the effect of missense changes on protein structure and function do not agree on the potential impact of this missense change (SIFT: "Deleterious"; PolyPhen-2: "Benign"; Align-GVGD: "Class C0"). This variant is present in population databases (rs779052721, ExAC 0.009%) but has not been reported in the literature in individuals with a TGFBR2-related disease. This sequence change replaces valine with alanine at codon 167 of the TGFBR2 protein (p.Val167Ala). The valine residue is highly conserved and there is a small physicochemical difference between valine and alanine. -

The valine at codon 167 is replaced by alanine, an amino acid with similar properties. Based on data from ExAC, the C allele has an overall frequency of less than 0.01% (1/106174). This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -
