The FBN2 c.3565C>T variant is predicted to result in the amino acid substitution p.His1189Tyr. This variant has been reported as a variant of uncertain significance in an individual with incomplete classical Ehlers–Danlos syndrome, who was later found to have an additional causative variant in COL5A1 gene (Ritelli et al. 2020. PubMed ID: 32720758). This variant is reported in 0.0046% of alleles in individuals of European (Non-Finnish) descent in gnomAD. At this time, the clinical significance of this variant is uncertain due to the absence of conclusive functional and genetic evidence.

This sequence change replaces histidine, which is basic and polar, with tyrosine, which is neutral and polar, at codon 1189 of the FBN2 protein (p.His1189Tyr). This variant is present in population databases (rs779690646, gnomAD 0.004%). This variant has not been reported in the literature in individuals affected with FBN2-related conditions. ClinVar contains an entry for this variant (Variation ID: 213399). Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is not expected to disrupt FBN2 protein function with a negative predictive value of 80%.
