The variant allele was found at a frequency of 4e-06 in 249786 control chromosomes (gnomAD). c.424C>T has been reported in two compound heterozygous individuals from one family affected with developmental delay, poor growth, and sensorineural hearing loss (Webb_2015). These data indicate that the variant may be associated with disease. One submitter has provided clinical-significance assessments for this variant to ClinVar after 2014 and classified the variant as pathogenic.
