The encoded protein is one of the components of a calcium channel, composed of a tetramer of the ryanodine receptor proteins and a tetramer of FK506 binding protein 1B proteins, that supplies calcium to cardiac muscle. This missense change has been observed in individuals with autosomal dominant catecholaminergic polymorphic ventricular tachycardia (PMID: 12093772, 19926015, 21292648, 23595086, 26114861). It has also been observed to segregate with disease in related individuals. ClinVar contains an entry for this variant (Variation ID: 201357). Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is expected to disrupt RYR2 protein function with a positive predictive value of 95%. For these reasons, this variant has been classified as Pathogenic. -

This variant has been detected in individuals with known or suspected catecholaminergic polymorphic ventricular tachycardia, and has been reported as occurring de novo in two cases (Priori SG et al.
