The STXBP1 c.1216C>A variant is predicted to result in the amino acid substitution p.Arg406Ser. To our knowledge, this variant has not been reported in the literature or in a large population database (http://gnomad.broadinstitute.org), indicating this variant is rare. De novo missense variants affecting the same amino acid residue have been reported in patients with epileptic encephalopathy (for example see Benson. 2020. PubMed ID: 32238909; Allen. 2016. PubMed ID: 26648591). Taken together, the c.1216C>A variant is interpreted as likely pathogenic. -
