Specifically, this protein functions as a member of a multi-protein complex that binds to the TTAGGG repeats of telomeres, regulating telomere length and protecting chromosome ends from illegitimate recombination, catastrophic chromosome instability, and abnormal chromosome segregation. Increased transcriptional expression of this gene is associated with stomach carcinogenesis and its progression. Alternatively spliced transcript variants have been described. This sequence change replaces glycine, which is neutral and non-polar, with cysteine, which is neutral and slightly polar, at codon 95 of the POT1 protein (p.Gly95Cys). This variant is not present in population databases (gnomAD no frequency). This missense change has been observed in individual(s) with glioma (PMID: 25482530). It has also been observed to segregate with disease in related individuals. ClinVar contains an entry for this variant (Variation ID: 209093). Invitae Evidence Modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) indicates that this missense variant is not expected to disrupt POT1 protein function with a negative predictive value of 80%. Experimental studies have shown that this missense change affects POT1 function (PMID: 33782098). In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. -

The p.G95C variant (also known as c.283G>T), located in coding exon 4 of the POT1 gene, results from a G to T substitution at nucleotide position 283. The glycine at codon 95 is replaced by cysteine, an amino acid with highly dissimilar properties. This variant was identified in one or more individuals with gliomas and segregated with disease in at least one family (Bainbridge MN et al. J Natl Cancer Inst, 2015 Jan;107:384). This amino acid position is well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear. -
