Ataxia-telangiectasia syndrome Uncertain:2
In summary, this is a novel in-frame deletion with uncertain impact on protein function. It has been classified as a Variant of Uncertain Significance. Experimental studies and prediction algorithms are not available for this variant, and the functional significance of the deleted amino acid is currently unknown. This variant is not present in population databases (ExAC no frequency) and has not been reported in the literature in individuals with an ATM-related disease. This sequence change deletes 3 nucleotides from exon 17 of the ATM mRNA (c.2589_2591delTAG). This leads to the deletion of 1 amino acid residue in the ATM protein (p.Ser864del) but otherwise preserves the integrity of the reading frame. -

Hereditary cancer-predisposing syndrome Uncertain:1
The c.2593_2595delAGT variant (also known as p.S865del) is located in coding exon 16 of the ATM gene. This variant results from an in-frame AGT deletion at nucleotide positions 2593 to 2595. This results in the in-frame deletion of a serine at codon 865. This amino acid position is not well conserved in available vertebrate species. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -
