This alteration has been identified in high-risk breast and ovarian cancer cohorts (Ramus SJ et al. J. Natl. Cancer Inst. 2015 Nov;107(11); Li J et al. J. Med. Genet., 2016 Jan;53:34-42). This variant is considered to be rare based on population cohorts in the Genome Aggregation Database (gnomAD). In addition to the clinical data presented in the literature, this alteration is expected to result in loss of function by premature protein truncation or nonsense-mediated mRNA decay. As such, this alteration is interpreted as a disease-causing mutation. -

The variant allele was found at a frequency of 4.3e-06 in 233042 control chromosomes (gnomAD). c.627_628delAA has been reported in the literature in multiple individuals affected with breast- or ovarian cancer (examples: Ramus_2015, Li_2016, Thompson_2016, and Dorling_2021). These data indicate that the variant is likely associated with disease. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported. Four clinical diagnostic laboratories have submitted clinical-significance assessments for this variant to ClinVar after 2014 and classified the variant as pathogenic (n=3) / likely pathogenic (n=1). Based on the evidence outlined above, the variant was classified as pathogenic. -
