The threonine at codon 613 is replaced by isoleucine, an amino acid with similar properties. This variant was detected in a patient with pancreatic adenocarcinoma and a history of cancer. This patient had a concurrent BRCA1 mutation and BRIP1 VUS (Dudley B et al. Cancer, 2018 04;124:1691-1700). This amino acid position is well conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -
