The p.T613I variant (also known as c.1838C>T), located in coding exon 12 of the BRIP1 gene, results from a C to T substitution at nucleotide position 1838. The threonine at codon 613 is replaced by isoleucine, an amino acid with similar properties. This variant was detected in a patient with pancreatic adenocarcinoma and a history of cancer. This patient had a concurrent BRCA1 mutation and BRIP1 VUS (Dudley B et al. Cancer, 2018 04;124:1691-1700). This amino acid position is well conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -
This missense variant replaces threonine with isoleucine at codon 613 of the BRIP1 protein. Computational prediction is inconclusive regarding the impact of this variant on protein structure and function (internally defined REVEL score threshold 0.5 < inconclusive < 0.7, PMID: 27666373). To our knowledge, functional studies have not been reported for this variant. This variant has been reported in an individual affected with ovarian cancer and pancreatic cancer who carried a pathogenic BRCA1 variant that could explain the observed phenotype (PMID: 29360161). This variant has not been identified in the general population by the Genome Aggregation Database (gnomAD). The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance. -
