This protein is an important cell cycle checkpoint kinase that phosphorylates; thus, it functions as a regulator of a wide variety of downstream proteins, including tumor suppressor proteins p53 and BRCA1, checkpoint kinase CHK2, checkpoint proteins RAD17 and RAD9, and DNA repair protein NBS1. This protein and the closely related kinase ATR are thought to be master controllers of cell cycle checkpoint signaling pathways that are required for cell response to DNA damage and for genome stability. The c.1095_1101delGATTTCTinsA variant, located in coding exon 8 of the ATM gene, results from an in-frame deletion of GATTTCT and insertion of A at nucleotide positions 1095 to 1101. This results in the in-frame deletion of the isoleucine and serine residues at codons 366 and 367 (p.I366_S367del). This amino acid region is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive (Choi Y et al. PLoS ONE. 2012; 7(10):e46688). Based on the available evidence, the clinical significance of this variant remains unclear. -
