This variant disrupts the p.Asp522 amino acid residue in TGFBR2. Other variant(s) that disrupt this residue have been observed in individuals with TGFBR2-related conditions (PMID: 17599521), which suggests that this may be a clinically significant amino acid residue. This sequence change replaces aspartic acid with alanine at codon 522 of the TGFBR2 protein (p.Asp522Ala). The aspartic acid residue is highly conserved and there is a moderate physicochemical difference between aspartic acid and alanine. This variant is not present in population databases (ExAC no frequency). This variant has been observed in a family with clinical features of TGFBR2-related disease (Invitae). ClinVar contains an entry for this variant (Variation ID: 408440). Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Deleterious"; PolyPhen-2: "Probably Damaging"; Align-GVGD: "Class C0").
