Five of five in-silico tools predict a benign effect of the variant on protein function. The variant was absent in 250604 control chromosomes (gnomAD). The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. c.928T>G has been reported in the literature as a VUS in an individual affected with triple negative breast cancer and a family history of lung, CNS and pancreatic cancers who underwent multigene panel testing (Tung_2016). This individual also had a co-occurrencing pathogenic variant (NBN c.127C>T, p.Arg43X), providing supporting evidence for a benign role, although family members were not genetically tested. This report does not provide unequivocal conclusions about association of the variant with Breast Cancer. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported.
