The protein encoded by this gene is involved in transcription-coupled nucleotide excision repair and is an integral member of the basal transcription factor BTF2/TFIIH complex. The gene product has ATP-dependent DNA helicase activity and belongs to the RAD3/XPD subfamily of helicases. Defects in this gene can result in three different disorders, the cancer-prone syndrome xeroderma pigmentosum complementation group D, trichothiodystrophy, and Cockayne syndrome. Functional studies measuring ultraviolet sensitivity in XP6BE cells demonstrated that the cells expressing the p.Ser23del variant were much more sensitive to UV light than cells expressing the wild type, suggesting that this variant affects the function of ERCC2. Frequency information for this variant is not available in the 1000 Genomes Project, the Exome Sequencing Project or the Exome Aggregation Consortium. The evidence for this variant is limited. The p.Ser23del variant is therefore classified as a variant of unknown significance but suspicious for pathogenicity for ERCC2-related disorders. This variant was observed by ICSL as part of a predisposition screen in an ostensibly healthy population. - |

Experimental studies and prediction algorithms are not available or were not evaluated, and the functional significance of this variant is currently unknown.
