The protein binds to DNA as either a homodimer, or a heterodimer with the related protein hepatocyte nuclear factor 1-alpha. The gene has been shown to function in nephron development, and regulates development of the embryonic pancreas. Mutations in this gene result in renal cysts and diabetes syndrome and noninsulin-dependent diabetes mellitus, and expression of this gene is altered in some types of cancer. This variant was reported in an individual with congenital or cystic renal disease (Supp. Table 7 in Groopman et al 2019. PubMed ID: 30586318). A different substitution at the same amino acid (p.Gly285Asp) has been reported in an individual with maturity-onset diabetes of the young type 5 (MODY 5) (Bellanne-Chantelot et al. 2005. PubMed ID: 16249435). This variant has not been reported in a large population database, indicating this variant is rare. Although we suspect that this variant may be pathogenic, at this time, the clinical significance of this variant is uncertain due to the absence of conclusive functional and genetic evidence. -
