The encoded protein functions as a calcium release channel in the sarcoplasmic reticulum but also serves to connect the sarcoplasmic reticulum and transverse tubule. Mutations in this gene are associated with malignant hyperthermia susceptibility, central core disease, and minicore myopathy with external ophthalmoplegia. This sequence change replaces glycine, which is neutral and non-polar, with serine, which is neutral and polar, at codon 1049 of the RYR1 protein (p.Gly1049Ser). The frequency data for this variant in the population databases is considered unreliable, as metrics indicate poor data quality at this position in the gnomAD database. This missense change has been observed in individual(s) with myopathy (PMID: 22473935, 25960145). ClinVar contains an entry for this variant (Variation ID: 544388).
