FBN1 (HGNC:3603): (fibrillin 1) This gene encodes a member of the fibrillin family of proteins. The encoded preproprotein is proteolytically processed to generate two proteins including the extracellular matrix component fibrillin-1 and the protein hormone asprosin. Fibrillin-1 is an extracellular matrix glycoprotein that serves as a structural component of calcium-binding microfibrils. These microfibrils provide force-bearing structural support in elastic and nonelastic connective tissue throughout the body. Asprosin, secreted by white adipose tissue, has been shown to regulate glucose homeostasis. Mutations in this gene are associated with Marfan syndrome and the related MASS phenotype, as well as ectopia lentis syndrome, Weill-Marchesani syndrome, Shprintzen-Goldberg syndrome and neonatal progeroid syndrome. Frameshifts are typically deleterious in nature; however, this frameshift occurs at the 3' terminus of FBN1, is not expected to trigger nonsense-mediated mRNA decay, and impacts only the last 69 amino acids of the C-terminal propeptide of the protein. Although the exact functional impact of these altered amino acids is unknown at this time, a functional study has demonstrated that other truncations within the C-terminal propeptide (both 5' and 3' of this alteration) interfere with FBN1 secretion (Jensen SA et al. Proc. Natl. Acad. Sci. U.S.A. 2014;111:10155-60). In addition, a number of truncations past this position have been reported in individuals with Marfan syndrome in the literature. This alteration is expected to result in loss of function by premature protein truncation. As such, this alteration is interpreted as a disease-causing mutation. -
