Variant summary: ATP8B1 c.923G>T (p.Gly308Val) results in a non-conservative amino acid change in the encoded protein sequence. Four of four in-silico tools predict a damaging effect of the variant on protein function. The variant was absent in 251404 control chromosomes (gnomAD). c.923G>T has been reported in the literature in several homozygous individuals affected with Familial Intrahepatic Cholestasis (e.g. van Wessel_2021). These data indicate that the variant is very likely to be associated with disease. Publication also reported experimental evidence evaluating an impact on protein function and demonstrated highly reduced protein stability, with the absence of plasma membrane expression (Folmer_2009), in addition, mice expressing G308V/G308V showed perturbed bile salt homeostasis (Pawlikowska_2004). The following publications have been ascertained in the context of this evaluation (PMID: 19731236, 14976163, 33666275). ClinVar contains an entry for this variant (Variation ID: 7262).
