The Q2109X pathogenic variant in the FBN1 gene has been previously published in individuals who met the Ghent criteria for Marfan syndrome (Collod-Béroud et al., 2003; Stheneur et al., 2009; Baetens et al., 2011). Q2109X is predicted to cause loss of normal protein function either by protein truncation or nonsense-mediated mRNA decay. Other downstream nonsense variants in the FBN1 gene have been reported in Human Gene Mutation Database in association with Marfan syndrome (Stenson et al., 2014). Furthermore, the Q2109X variant is not observed in large population cohorts (Lek et al., 2016).In summary, Q2109X in the FBN1 gene is interpreted as a pathogenic variant. -

This sequence change creates a premature translational stop signal (p.Gln2109*) in the FBN1 gene. It is expected to result in an absent or disrupted protein product. Loss-of-function variants in FBN1 are known to be pathogenic (PMID: 17657824, 19293843). This variant is not present in population databases (gnomAD no frequency). This premature translational stop signal has been observed in individual(s) with Marfan syndrome (PMID: 19293843, 21542060). In at least one individual the variant was observed to be de novo. ClinVar contains an entry for this variant (Variation ID: 488820). For these reasons, this variant has been classified as Pathogenic. -
