Genes affected
FBN1 (HGNC:3603): (fibrillin 1) This gene encodes a member of the fibrillin family of proteins. The encoded preproprotein is proteolytically processed to generate two proteins including the extracellular matrix component fibrillin-1 and the protein hormone asprosin. Fibrillin-1 is an extracellular matrix glycoprotein that serves as a structural component of calcium-binding microfibrils. These microfibrils provide force-bearing structural support in elastic and nonelastic connective tissue throughout the body. Asprosin, secreted by white adipose tissue, has been shown to regulate glucose homeostasis. Mutations in this gene are associated with Marfan syndrome and the related MASS phenotype, as well as ectopia lentis syndrome, Weill-Marchesani syndrome, Shprintzen-Goldberg syndrome and neonatal progeroid syndrome. Pathogenic, criteria provided, single submitterclinical testingGeneDxDec 20, 2017The Q2109X pathogenic variant in the FBN1 gene has been previously published in individuals who met the Ghent criteria for Marfan syndrome (Collod-Béroud et al., 2003; Stheneur et al., 2009; Baetens et al., 2011). Q2109X is predicted to cause loss of normal protein function either by protein truncation or nonsense-mediated mRNA decay. Other downstream nonsense variants in the FBN1 gene have been reported in Human Gene Mutation Database in association with Marfan syndrome (Stenson et al., 2014). Furthermore, the Q2109X variant is not observed in large population cohorts (Lek et al., 2016).In summary, Q2109X in the FBN1 gene is interpreted as a pathogenic variant. -
Marfan syndrome;C4707243:Familial thoracic aortic aneurysm and aortic dissection Pathogenic:1
Pathogenic, criteria provided, single submitterclinical testingInvitaeAug 12, 2018This variant is not present in population databases (ExAC no frequency). For these reasons, this variant has been classified as Pathogenic. Loss-of-function variants in FBN1 are known to be pathogenic (PMID: 17657824, 19293843). This variant has been observed in individuals affected with Marfan syndrome, having been reported to be de novo in one of them (PMID: 19293843, 21542060). ClinVar contains an entry for this variant (Variation ID: 488820). This sequence change creates a premature translational stop signal (p.Gln2109*) in the FBN1 gene. It is expected to result in an absent or disrupted protein product. -
