Pathogenic, criteria provided, single submitter | clinical testing | Ambry Genetics | Apr 20, 2015 | The p.C1942S pathogenic mutation (also known as c.5825G>C), located in coding exon 47 of the FBN1 gene, results from a G to C substitution at nucleotide position 5825. The cysteine at codon 1942 is replaced by serine, an amino acid with dissimilar properties, and is located in the cb EGF-like #29 domain. The majority of FBN1 mutations identified to date have involved the substitution or generation of cysteine residues within cbEGF domains (Vollbrandt T et al. J Biol Chem. 2004;279(31):32924-32931). This alteration has been determined to be the result of a de novo mutation or germline mosaicism in one family with Marfan syndrome. Based on the supporting evidence, p.C1942S is interpreted as a disease-causing mutation. - |
