This receptor/ligand complex phosphorylates proteins, which then enter the nucleus and regulate the transcription of genes related to cell proliferation, cell cycle arrest, wound healing, immunosuppression, and tumorigenesis. Mutations in this gene have been associated with Marfan Syndrome, Loeys-Deitz Aortic Aneurysm Syndrome, and the development of various types of tumors. The p.G509D variant (also known as c.1526G>A), located in coding exon 7 of the TGFBR2 gene, results from a G to A substitution at nucleotide position 1526. The glycine at codon 509 is replaced by aspartic acid, an amino acid with similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on internal structural analysis, G509D is more destabilizing to the structure of TGFBR2 than several pathogenic variants in the same domain (Tebben AJ et al. Acta Crystallogr D Struct Biol, 2016 05;72:658-74; Reddy KB et al. Biochemistry, 1996 Jan;35:309-14). Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -
