Mutations in this gene cause Gaucher disease, a lysosomal storage disease characterized by an accumulation of glucocerebrosides. Variant summary: The GBA c.630delC (p.Val211Phefs) variant results in a premature termination codon, predicted to cause a truncated or absent GBA protein due to nonsense mediated decay, which are commonly known mechanisms for disease. Truncations downstream of this position have been classified as pathogenic by our laboratory (e.g.c.1029delT/ p.Tyr343fs). One in silico tool predicts a damaging outcome for this variant. This variant is absent in 74870 control chromosomes. However, it has been reported in multiple GD patients as compound heterozygotes (mostly with Korean ethnicities). Taken together, this variant is classified as pathogenic. -
