The p.H215Q variant (also known as c.645C>G), located in coding exon 4 of the TGFBR2 gene, results from a C to G substitution at nucleotide position 645. The histidine at codon 215 is replaced by glutamine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear. -
This sequence change replaces histidine, which is basic and polar, with glutamine, which is neutral and polar, at codon 215 of the TGFBR2 protein (p.His215Gln). This variant is present in population databases (rs758827786, gnomAD 0.003%). This variant has not been reported in the literature in individuals affected with TGFBR2-related conditions.
