The p.A440S variant (also known as c.1318G>T), located in coding exon 11 of the MRE11A gene, results from a G to T substitution at nucleotide position 1318. The alanine at codon 440 is replaced by serine, an amino acid with similar properties. This amino acid position is well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -

This sequence change replaces alanine, which is neutral and non-polar, with serine, which is neutral and polar, at codon 440 of the MRE11 protein (p.Ala440Ser). This variant is present in population databases (rs773469981, gnomAD 0.01%). This variant has not been reported in the literature in individuals affected with MRE11-related conditions. ClinVar contains an entry for this variant (Variation ID: 479776).
